RESUMO

Essential oil from Coriandrum sativum has been demonstrated to provide various pharmacological properties, such as antioxidant, antimicrobial, antibacterial, antifungal, antidiabetic, anticonvulsive, anxiolytic-antidepressant, and anti-aging properties. This study investigated the mechanism of Coriandrum sativum var. microcarpum essential oil (CSEO, 25, 150, and 300 µL/L) and cognitive impairment and brain oxidative stress in a scopolamine (SCOP, 100 µM) zebrafish model (Danio rerio) of cognitive impairment. Spatial memory, response to novelty, and recognition memory were assessed using the Y-maze test and the novel object recognition test (NOR), while anxiety-like behavior was investigated using the novel tank diving test (NTT). The cholinergic system activity and brain oxidative stress were also evaluated. CSEO was administered to zebrafish once a day for 21 days, while SCOP and galantamine (GAL, 1 mg/L) were delivered 30 min before behavioral testing and euthanasia. Our data revealed that SCOP induced memory dysfunction and anxiety-like behavior, while CSEO improved memory performance, as evidenced by behavioral tasks. Moreover, CSEO attenuated SCOP-induced brain oxidative stress and decreased acetylcholinesterase (AChE) activity. The results demonstrated the potential use of the CSEO in providing beneficial effects by reducing memory deficits and brain oxidative stress involved in the genesis of a dementia state.

With the increasing demand for European catfish, traditional extensive growth methods in polyculture are no longer sufficient to meet market needs. Therefore, this study aimed to identify indicators for improving recirculating aquaculture system (RAS) technology by determining and comparing growth performance, flesh quality, blood profile, oxidative status, and intestinal microbiota parameters between fish cultivated in a RAS and an earthen pond. Results revealed that RAS-grown fish had a higher fat content compared to pond-grown fish, while no significant differences were found for growth parameters. Sensory analysis showed no significant difference in taste between the two groups. Blood composition analysis showed small differences. Oxidative status analyses showed higher catalase and glutathione peroxidase activities in RAS-grown fish and slightly higher superoxide dismutase activity in pond-grown fish. Microbial analysis showed differences in the intestinal microflora, with a higher total number of aerobic germs and anaerobic germs and a lower total number of sulfite-reducing clostridia in RAS-grown fish. This study provides valuable insights into the comparative performance of a RAS and a pond rearing system in European catfish production, potentially informing future growth technologies.

The purpose of this study was to investigate the effect of Glaucosciadium cordifolium essential oil (GCEO, 25 and 150 µL/L) on anxiety and learning and memory impairment induced by scopolamine (SCOP) in zebrafish. The chemical composition was analyzed by GC-MS, and the results showed that the highest content was limonene followed by α- and ß-pinene, p-cymene and α-phellandrene. The dementia model was induced by SCOP (100 µM), whereas GCEO and galantamine (GAL, 1 mg/L) were delivered to the SCOP-induced model. It was found that GCEO significantly improved memory impairment and anxiety-like response induced by SCOP through the Y-maze, novel object recognition (NOR) test, and novel tank diving tests (NTT). Biochemical analyses showed that GCEO reduced SCOP-induced oxidative damage. Additionally, the cholinergic system activity was improved in the SCOP-induced model by decreasing the acetylcholinesterase (AChE) activity following the exposure to GCEO. It was clear that as a mixture, GCEO displays positive action in improving memory impairment through restoring cholinergic dysfunction and brain antioxidant status.

Distillers dried grains with solubles (DDGS), a coproduct from the ethanol production industry, is successfully used as an ingredient in feeding cattle and pigs due to its relatively high protein and nutrient content and low price compared to cereals. The aim of this study was to establish the optimal DDGS concentration that can be included in the diet of common carp. A seven-week experiment was performed on common carp with an initial weight of 86 g feed with three experimental diets D0 (DDGS 0%), D1 (DDGS 25%) and D2 (DDGS 35%). The chemical composition of DDGS analyzed by Fourier Transform Near-Infrared (FT-NIR) spectroscopy showed a protein content of 27.56% and oil at 6.75%. Diets with DDGS did not produce significant changes in growth parameters, flesh quality, and blood biochemical profile. Regarding the oxidative status in the muscle tissue, D1 and D2 significantly reduced, in a dose-dependent manner, the specific activity of SOD and GSH, while CAT and GPX were left unaffected. In the liver tissue, CAT, GSH, MDA and carbonylated proteins were reduced in the DDGS diets. The microbiological analysis of the intestinal contents revealed a variation in microbial density depending on the diet used. The total number of aerobic germs was between 224.2 × 104 and 69.84 × 106 (D2 > D1 > D0) and the total number of anaerobic germs was between 15.2 × 102 and 28.2 × 102 (D2 > D0 > D1).

Angelica purpurascens (Avé-Lall.) Gilli. is a medicinal plant that displays antioxidant, anticholinesterase, and neuroprotective properties. The effect of A. purpurascens essential oil (APO) on memory impairments and brain oxidative stress in zebrafish (Danio rerio) treated with scopolamine (Sco), as well as the underlying mechanism involved, were investigated in this study. Exposure to Sco (100 µM) resulted in anxiety in zebrafish, as assessed by the novel tank diving test (NTT), whereas spatial memory and novelty response dysfunctions, as evidenced by the Y-maze test and novel object recognition test (NOR), were noticed. When zebrafish were given Sco and simultaneously given APO (25 and 150 µL/L, once daily for 13 days), the deficits were averted. An increase in brain antioxidant enzymes, a reduction of lipid peroxidation, and protein oxidation were linked to this impact. Furthermore, acetylcholinesterase (AChE) activity was significantly reduced in the brains of APO-treated zebrafish. The main detected components in the APO composition were ß-phellandrene (33.80%), sabinene (6.80%), α-pinene (5.30%), germacrene-D (4.50%), α-phellandrene (4.20%), and p-cymene (3.80%) based on gas chromatography-mass spectrometry (GC-MS) investigations. Our findings show that APO's beneficial effect in a zebrafish model of Sco-induced memory impairment is mediated through multiple mechanisms, including the restoration of cholinergic system function and the improvement of the brain antioxidant state. As a result, APO could be employed as a potential source of bioactive molecules with useful biological properties and medicinal uses.

Thymus vulgaris L. is an aromatic herb used for medicinal purposes such as antimicrobial, spasmolytic, antioxidant, anti-inflammatory, antinociceptive, antitumor, and may have beneficial effects in the treatment of Alzheimer's disease. The present study aimed to investigate whether Thymus vulgaris L. essential oil enhances cognitive function via the action on cholinergic neurons using scopolamine (Sco)-induced zebrafish (Danio rerio) model of memory impairments. Thymus vulgaris L. essential oil (TEO, 25, 150, and 300 µL/L) was administered by immersion to zebrafish once daily for 13 days, whereas memory impairment was induced by Sco (100 µM), a muscarinic receptor antagonist, delivered 30 min before behavioral tests. Spatial memory was assessed using the Y-maze test and novel object recognition test (NOR). Anxiety and depression were measured in the novel tank diving test (NTT). Gas Chromatograph-Mass Spectrometry (GC-MS) analysis was used to study the phytochemical composition of TEO. Acetylcholinesterase (AChE) activity and oxidative stress response in the brain of zebrafish were determined. TEO ameliorated Sco-induced increasing of AChE activity, amnesia, anxiety, and reduced the brain antioxidant capacity. These results suggest that TEO may have preventive and/or therapeutic potentials in the management of memory deficits and brain oxidative stress in zebrafish with amnesia.

Ceratonia siliqua L. is a Mediterranean medicinal plant traditionally cultivated for its ethnopharmacological benefits, such as antidiarrheal, antidiabetic, enhance acetylcholine, antioxidant, antiatherosclerotic, and for its possible anti-neurodegenerative potential. The aim of the present study was to evaluate the chemical composition, as well as the cognitive-enhancing, anxiolytic, and antioxidant activities of the aqueous extract from C. siliqua (CsAE) leaves against 6-hydroxydopamine (6-OHDA) zebrafish Parkinson's disease (PD) model. CsAE (0.1, 0.3, and 1 mg/L) was administered by immersion to zebrafish (Danio rerio) for eight consecutive days and one hour before each behavioral test of each day, while 6-OHDA (250 µM) treatment was supplied one day before the novel tank diving test (NTT). Qualitative and quantitative analyses were performed by the ultra-high-performance liquid chromatography (UHPLC) analysis. The memory performance was evaluated through the NTT and Y-maze tests. Additionally, the in vitro and in vivo antioxidant status and acetylcholinesterase (AChE) activity was also assessed. Our finds demonstrated that CsAE presented positive antioxidant and anti-AChE activities, which contributed to the improvement of cognitive function in the 6-OHDA zebrafish PD model.

Rosmarinus officinalis L. is a traditional herb with various therapeutic applications such as antibacterial, antioxidant, anti-inflammatory, antidepressant, and anticholinesterase activities, and can be used for the prevention or treatment of dementia. In the present study, we tested whether Rosmarinus officinalis L. could counteract scopolamine-induced anxiety, dementia, and brain oxidative stress in the zebrafish model and tried to find the underlying mechanism. Rosmarinus officinalis L. essential oil (REO: 25, 150, and 300 µL/L) was administered by immersion to zebrafish (Danio rerio) once daily for eight days while scopolamine (100 µM) treatment was delivered 30 min before behavioral tests. The antidepressant and cognitive-enhancing actions of the essential oil in the scopolamine zebrafish model was measured in the novel tank diving test (NTT) and Y-maze test. The chemical composition was identified by Gas chromatograph-Mass spectrometry (GC-MS) analysis. The brain oxidative status and acetylcholinesterase (AChE) activity was also determined. REO reversed scopolamine-induced anxiety, memory impairment, and brain oxidative stress. In addition, a reduced brain AChE activity following the administration of REO in scopolamine-treated fish was observed. In conclusion, REO exerted antidepressant-like effect and cognitive-enhancing action and was able to abolish AChE alteration and brain oxidative stress induced by scopolamine.

Schinus terebinthifolius is a plant well recognized for its therapeutic profile such as anti-inflammatory and antitumor activities, promoting antibacterial activity and antioxidant and antidiabetic properties. This study aimed at examining whether Schinus terebinthifolius memory-enhancing activities are mediated by cholinergic and brain antioxidant systems in a scopolamine zebrafish model. Schinus terebinthifolius essential oil (10, 25, and 50 µL/L) was delivered to zebrafish by immersion in water for 8 days. Memory deficits were induced by scopolamine (100 µM) administration. Zebrafish were divided into seven groups (n = 15/group): vehicle group, scopolamine (100 µM) group, Schinus terebinthifolius essential oil groups (STF; 10, 25, and 50 µL/L), the imipramine group (IMP; 20 mg/L, as the positive control in the NTT test), and the donepezil group (DP; 10 mg/L, as the positive control in the Y-maze test). Memory status was estimated by the novel tank diving test (NTT) and the Y-maze test and finally was validated by comparison with imipramine (20 mg/L) and donepezil (10 mg/L). Gas chromatography-mass spectrometry (GC-MS) was used to detect oil compounds. Brain levels of acetylcholinesterase (AChE) and antioxidant enzymes were measured. After being exposed to Schinus terebinthifolius essential oil, the scopolamine zebrafish exhibited an improvement of memory processes in the NTT and Y-maze tests. The essential oil attenuated the elevated level of AChE and brain oxidative stress. Schinus terebinthifolius essential oil was found to support memory formation through the inhibition of the AChE activity and decreasing oxidative stress in the scopolamine-treated zebrafish brains.

BACKGROUND: Agathisflavone, a biflavonoid isolated from Schinus polygamus (Cav.) Cabrera leaves been reported to promote various biological activities such as anti-inflammatory properties, promoting cognition and preventing cancer, antioxidant and antiapoptotic activities. PURPOSE: Here, we tested the hypothesis whether anxiety, amnesia, and brain oxidative stress induced by scopolamine could be counteracted in zebrafish model by agathisflavone and tried to ascertain the underlying mechanism. METHODS: Agathisflavone (1, 3 and 5 µg/l) was administered by immersion to zebrafish once daily for 8 days period. Anxiety and memory impairment were induced with scopolamine (100 µM) and measured with the novel tank diving test (NTT) and the Y-maze test. Zebrafish were divided into seven groups (n = 20/group): first group - control, second group - scopolamine (100 µM), the third, fourth and fifth group - agathisflavone treatment groups (FAB, 1 µg/l, 3 µg/l, and 5 µg/l), the sixth group - imipramine (IMP, 20 mg/l, as the positive control in NTT test), and the seventh group - donepezil group (DP, 10 mg/l, as the positive control in Y-maze test). The identification of the agathisflavone was done by spectroscopy, and the structure of the compound was confirmed by (-) Electrospray Ionisation Mass Spectrometry (ESI-MS). The brain oxidative status and acetylcholinesterase (AChE) activity were also investigated. RESULTS: Agathisflavone from Schinus polygamus (Cav.) Cabrera leaves was identified. Also, we demonstrated that agathisflavone significantly reversed scopolamine-induced behavioral score alteration in the NTT and Y-maze tests. Consequently, agathisflavone promoted inhibition of AChE activity and restored the brain antioxidant status. CONCLUSION: Our results demonstrate that agathisflavone promotes brain antioxidant action and ameliorates scopolamine-induced anxiety and memory deficits in zebrafish.

Abstract The accumulation of heavy metals and of pesticides in the soil have a negative impact on Capsicum chinense var. Aji Mochero and Capsicum baccatum var. pendulum Bishop's Crown species, the present work highlighting the exerted influence on the superoxid-dismutase activity, catalase and peroxidase - as enzymes implied in the removal of H2O2, of O2- radical and of ion HO- that have devastating effects on the vegetable cell, on the Krebs cycle's dehydrogenases - as main way to produce energy, respectively of the chlorophyll a, b and carotenes - as photoassimilatory pigments, but also as biologic antioxidants.

Gold is a noble metal having a long history of human usage and a variety of applications. The present paper was designed in order to see the changes on social and swimming behavior of zebrafish caused by acute gold exposure to high concentrations dissolved in aquatic medium from a standard solution with the highest purity. Some main oxidative stress markers were determined such as: superoxide dismutase (SOD), catalase (CAT), malondialdehyde (MDA) and glutathione peroxidase (GPx). We also focused our attention on the bioaccumulation capacity of gold in exposed zebrafish and its competition with essential elements for the body: sodium (Na), potassium (K), calcium (Ca), magnesium (Mg), iron (Fe), zinc (Zn) and copper (Cu). They were studied the effects of the follow gold concentrations dissolved in experimental aquariums: 1 mg L-1 (Au 1 mg/L), 2 mg L-1 (Au 2 mg/L), 4 mg L-1 (Au 4 mg/L) and 8 mg L-1 (Au 8 mg/L). Our data showed that each group treated with gold had a higher concentration compared with the others suggesting that it can be absorbed into the zebrafish body from environment and may be accumulated. The results obtained in the oxidative stress parameters demonstrated that it can produce changes as a response to its toxicological effects.

Fishes are the first group of vertebrates that respond when the environment is contaminated with pollutants resulted from anthropogenic activities. The development of the toxicity tests is bringing new evidence about the toxicological effects of the pollutants upon the life forms. Behavioural abnormalities in the swimming performance and cognitive processes were well associated with the response of organisms to pollutants from environment. The aim of the paper was to study the behavioural changes of zebrafish (memory, swimming performances and aggression) and oxidative stress (superoxide dismutase and malondialdehyde) during 32 h of acute exposure with methylmercury (II) chloride to measure its neurotoxicity effects upon fish community. The experiments from this study tested and measured the fish community response to methylmercury concentrations (1 µg L-1 and 15 µg L-1) in the first hours after it contamination based on zebrafish model. The changes of the behaviour in the case of a fish species may lead in the end to their population reduction based on less reproductive success, lower food resource exploitation and problems in the predator avoidance. The behavioural tests described in the present study can be applied to measure the neurotoxicity of other metals compounds, to do plans and protocols for avoiding future ecological disasters. The behavioural changes of zebrafish exposed to methylmercury (II) chloride were similar to mammal models and they will have applications in future research.

ABSTRACT It is known the fact that the mining causes huge damages on the environment having negative biologic effects extremely severe on some groups of organisms of the food chain. The pollutants causes major damages at the level of leaves' cuticles, the vegetation reacting through irreversible modifications at the tissue's level and at substances and energy metabolism, like a riposte to the apparition of the stress induced by the ROS, stress that creates real disturbances on physiological and biochemical processes. Considering the fact that, on the one hand, the activity of oxidoreductases is strongly influenced by the presence of the pollutants factors, with direct effect on the oxidative's stress production, and on the other hand that the plants react at ROS accumulation, through self-protection and adaptability mechanisms, the aim of this study consisted in monitoring the activity of some oxidoreductases in the leaves of plantain and mint species harvested in 2015 from different mining areas of Suceava County. In parallel was determined the dehydrogenase activity of the soil, the samples being collected from the same areas subdued to anthropic pollution, using like reference swatch, samples from Putna. The obtained results signalize the accumulation of some quantities highly arisen by free radicals of oxygen at investigated species, the enzymatic activity being significantly higher in the mining plant comparatively to the control areal. The microbial dehydrogenases from the soil samples were remarked through different activities based on the types of heavy metals that pollute each harvesting area and on the deepness profile.

Oxidative stress can be regarded as an imbalance between the amount of reactive oxygen species (ROS) and the ability of a biological system to eliminate the toxic species and repair the resulting damages. Since the germinating seeds and the resulted seedlings are rich in enzymes, whereas the treatment with chemicals affects much the seed germination, producing also ROS, we evaluate here the influence of 2,4-dinitrophenol (DNP) and potassium iodate (KIO3) on wheat germination (Triticum aestivum L.) and seedlings growth. Germination rate, the masses and heights of the 7 day old seedlings, as well as the activity of some enzymes involved in the oxidative stress such as peroxidase, catalase and superoxide dismutase were measured seven days after the chemical treatment. The treatment of the wheat seeds with 10-5 - 10-3 M solutions of DNP resulted in a relative concentration-dependent inhibition of the germination, with a concomitant stimulation of the weight and height of viable seedlings. The Gasparom variety treated with 10-5 M KIO3 showed a slight increase in the germination rate in comparison with the control batch. The two tested substances determined a significantly modified response of the oxidative stress enzymes, especially in the seeds treated with 10-4 and 10-3 M solutions.
